Isolation of DNA From Mouse Tails
Protocol 12.3B from Manipulating the Mouse Embryo 3rd Edition, Andras Nagy et al.

1. Place the tail sample (1-2mm) in 200 - 300 ul Lysis Buffer containing Proteinase K (Recipe below). Samples can be stored frozen until use.
2. Incubate the samples at 55°C for 3-4 hours to overnight. The DNA lysates can now be stored at 4°C if desired.
3. Optional: Centrifuge for 2-3 minutes to pellet hair and debris.
4. Incubate the samples at 95°C for 10 minutes before starting PCR amplification to inactivate Proteinase K.
5. Use 1-2ul lysate for each PCR.

Lysate Buffer:
50 mM KCl
10 mM Tris-HCl
2 mM MgCl2
0.1 mg/ ml gelatin
0.45% Nonidet P40
0.45% Tween 20
Aliquots (1 ml) are stored at -20°C.
Proteinase K, 500 ug/ ml, added just before use (stock 20 mg/ ml)
